Repid, Simple Method for Extraction of Mycobacteria Species DNA for Polymerase Chain Reaction

Abstract

|
|
A new simplified and effective method for detecting mycobacteria by the polymerase chain reaction(PCR) was investigated. The method is based on lysing and nuclease-inactivating properties of the chaotropic agent guanidinium thiocyanate(GuSCN)
together
with the DNA-binding properties of silica particles. We have evaluated the sensitivity of this method with 50 clinical samples from patients suspected of having tuberculosis and compared with conventional procedures for DNA purification.

detergent mediated lysis, proteinase treatment, extaction with organic solvents, and ethanol precipitation). In our opinion new method is more sensitive, rapid and reliable than that of conventional method for detetion of mycobacterial DNA in
clinical
specimens by PCR.
